Extra Details:
ATP7b is an important protein for copper transport and elimination of excess copper from the body. ATP7b transports metals in and out of cells using ATP. There are 3 known isoforms of the ATP7b gene; A is found in the liver, kidney, and brain, the shorter form B is found in brain tissue, and the third isoform, known as WND/140kD is found in mitochondria. Mutations in the ATP7b gene can cause Wilson’s disease, an inherited disorder causing copper poisoning in the brain and liver. Applications: Suitable for use in Western Blot, Immunocytochemistry and Immunohistochemistry. Other applications not tested. Recommended Dilution: Immunohistochemistry (Paraffin): 1:500 Western Blot: 1:1000. Band at ~165kD Immunocytochemistry: 1:500 Optimal dilutions to be determined by the researcher. Positive Control: HuH7 hepatocarcinoma cell extracts Cellular Localization: Golgi apparatus; Predominantly found in trans-Golgi network membrane; multi-pass membrane protein (By similarity). Isoform 2: Cytoplasm. The cleaved form WND/140kD is located in mitochondria. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
