- Applications: Suitable for use in Neutralization, ELISA and Western Blot. Other applications not tested. Recommended Dilutions: Western Blot: 0.1ug/ml using lysates of A549 human lung carcinoma cell line, HeLa human cervical epithelial carcinoma cell line, or HEK293T human embryonic kidney cell line Neutralization of receptor-ligand interaction: 0.5-2ug/ml Blocks 50% of the binding of 50ng/mL of rhCripto 1 to immobilized rhActRIB/Fc (100ul of 2ug/ml was coated in each well) in a functional ELISA. At 20ug/ml there will be >90% blocking KO Validation: Activin RIB/ALK‑4 is specifically detected in HeLa human cervical epithelial carcinoma parental cell line but is not detectable in Activin RIB/ALK‑4 knockout HeLa cell line. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
