- The cytoplasmic cytoskeleton determines cell organization, shape and adhesion among other functions. Furthermore, the cell type-specific expression of intermediate filaments allows determination of the origin of many, otherwise unspecific tumors. Applications: Suitable for use in Western Blot, Immunocytochemistry and Immunohistochemistry. Not suitable for use in Immunoprecipitation. Other applications have not been tested. Recommended Dilutions: Western Blot: 0.5-2ug/ml. Detects alpha-Actinin in RIPA lysates from human A431 cells. Immunocytochemistry/Immunohistochemistry: 15ug/ml. Reported positive immunostaining for alpha-Actinin in MRC-5 cells fixed with methanol and also in frozen sections. Optimal dilutions to be determined by the researcher. Positive Antigen Control (Included): A0001-50: A431 Cells, Non-Stimulated in RIPA Buffer, Sample Buffer. Recommended Secondary Antibodies: I1904-06C: IgG, H&L (HRP) (X-Adsorbed) Pab Gt x Mo I1904-06H: IgG, H&L (HRP) (X-Adsorbed) Pab Gt x Mo I1904-06F: IgG, H&L (FITC) (X-Adsorbed) Pab Gt x Mo I1904-23: IgG, X-Adsorbed (DTAF) Pab Gt x Mo Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
