Extra Details:
ABCG2, also called MXR, BCRP or ABCP, is highly overexpressed in several drug-resistant cell lines and localizes predominantly to the plasma membrane (Rocchi, E. et al., 2000, Biochem. Biophys. Res. Commun. 271:42-46). ABCG2 has also been shown to be expressed at relatively high levels in human hematopoietic stem cells (CD34+, CD38-) and to drop sharply in committed progenitors (CD34+/CD38+) (Scharenberg, C.W. et al., 2002, Blood 99:507-512). Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilution: Flow Cytometry: Designed to quantitatively determine the percentage of cells bearing Bcrp1/ABCG2 on their cell surface within a population and qualitatively determine the density of this receptor on cell surfaces by flow cytometry. Washed cells are incubated with the PE-labeled monoclonal antibody which binds to the cells expressing the Bcrp1/ABCG2 structure. Unbound PE-conjugated antibody is then washed from the cells. Cells expressing Bcrp1/ABCG2 become fluorescently stained, with the intensity of staining directly proportional to the density of the Bcrp1/ABCG2. Cell surface expression of Bcrp1/ABCG2 is determined by flow cytometric analysis using 488nm wavelength laser excitation. Storage and Stability: Store product at 4°C before and after reconstitution. DO NOT FREEZE! Lyophilized and reconstituted products are stable for 12 months at 4°C after receipt. Reconstitute lyophilized powder with sterile PBS prior to use and store remainder as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: PE conjugates are sensitive to light. For maximum recovery of product centrifuge the original vial prior to removing the cap.
