- The Stress-activated protein kinases alpha and gamma (SAPKalpha and gamma), also know as c-Jun NH2-terminal kinases 1 and 2 (Jnk1/2) are serine/ threonine protein kinases belonging to the MAP kinase family. The extracellular stimuli leading to cell stress include inflammatory cytokines (via Rho and Rac), growth factors, protein synthesis inhibitors and osmotic stress amongst others. Both Jnk1 and Jnk2 must be dually phosphorylated in the activation loop, at threonine 183 and tyrosine 185, to achieve full enzymatic activity. Applications: Suitable for use in ELISA, Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilutions: ELISA: 0.05ug/ml Western Blot (ECL): 0.5ug/ml Immunohistochemistry: 1:50 Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
