- Source: Recombinant protein corresponding to aa105-420 from human TPH1, fused to His-Tag at N-terminal, expressed in E.coli. Molecular Weight: ~37kD Applications: Suitable for use in the study of enzyme kinetics, screening inhibitors, and selectivity profiling. Other applications not tested. Assay Conditions: Reaction was performed at room temperature in assay buffer containing 50mM MOPS, pH 7, 60uM tryptophan, 100mM ammonium sulfate, 100uM ferrous ammonium sulfate, 0.5mM TCEP, 0.3mM 6-methyl tetrahydropterin, 0.05mg/ml catalase, 1mM DTT and TPH1. Fluorescence signal was measured at Em360nm/Ex300nm. Storage and Stability: Aliquot to avoid repeated freezing and thawing and store at -70°C. Aliquots are stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
