- Interleukin-6 (IL-6) is a pleiotropic cytokine produced by a variety of cells during infection, trauma, and immunological challenge (PMID: 8274730). IL-6 acts via a receptor complex consisting of two distinct membrane-bound glycoproteins, an 80kD IL-6-binding subunit (IL-6R, CD126, gp80) and a 130kD signal-transducing element (gp130, CD130). After binding of IL-6 to membrane-bound IL-6R, the complex of IL-6 and IL-6R associates with gp130, thus activating the receptor (PMID: 9716487). Expression of gp130 is found in almost all organs while expression of IL-6R is predominantly confined to hepatocytes and leukocyte subpopulations (monocytes, neutrophils, T cells, and B cells) (PMID: 11149892). Soluble form of IL-6R has been found in human serum and urine. Applications: Suitable for use in Western Blot, Immunohistochemistry and ELISA. Other applications not tested. Recommended Dilution: Western Blot: 1:500-1:2000, incubate membrane with diluted primary antibody in TBS, 5% milk, 0.1% Tween 20 at 4°C with gentle shaking, overnight. Immunohistochemistry: 1:50-1:200 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
