- BMAL1/2-CLOCK heterodimers activate E-box element (3'- CACGTG-5') transcription of a number of proteins of the circadian clock. Activates transcription of PER1 and PER2. This transcription is inhibited in a feedback loop by PER and CRY proteins. Component of the circadian clock oscillator which includes the CRY proteins, CLOCK or NPAS2, BMAL1 or BMAL2, CSNK1D and/or CSNK1E, TIMELESS and the PER proteins. Efficient DNA binding requires dimerization with another bHLH protein. Heterodimerization with BMAL1 is required for E-box-dependent transactivation, for CLOCK nuclear translocation and degradation, and, for phosphorylation of both CLOCK and BMAL1. Interaction with PER and CRY proteins requires translocation to the nucleus. Interaction of the CLOCK-BMAL1 heterodimer with PER or CRY inhibits transcription activation. Binds weakly ARNT and ARNT2 to form heterodimers which bind poorly to the E-box motif. Applications: Suitable for use in Western Blot, Immunocytochemistry and ELISA. Other applications not tested. Recommended Dilution: ELISA: 1:10,000 Western Blot: 1:500-1:2000 Immunocytochemistry: 1:200-1:1000 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
