Extra Details:: Recombinant monoclonal antibody to p53. Manufactured using Recombinant Platform with variable regions (i.e. specificity) from the hybridoma PAb421. p53, a DNA-binding, oligomerization domain and transcription activation domain-containing tumor suppressor, upregulates growth arrest and apoptosisrelated genes in response to stress signals, thereby influencing programmed cell death, cell differentiation and cell cycle control mechanisms. p53 localizes to the nucleus, yet can be chaperoned to the cytoplasm by the negative regulator MDM2, an E3 ubiquitin ligase that is upregulated in the presence of active p53, where MDM2 poly-ubiquitinates p53 for proteasome targeting. p53 fluctuates between latent and active (DNA-binding) conformations and is differentially activated through posttranslational modifications including phosphorylation and acetylation. Applications: Suitable for use in Immunofluorescence, ELISA, Western Blot, Immunoprecipitation, Immunohistochemistry and Immunohistochemistry. Other applications not tested. Recommended Dilution: Immunohistochemistry: Paraffin, In vivo Immunohistochemistry: Frozen Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
