- Extra Details:
- Proliferating cell nuclear antigen (PCNA) is a member of the DNA sliding clamp family of proteins that assist in DNA replication (1). Crystal structure data suggests that a PCNA homotrimer ring can encircle and slide along the DNA double helix (2). Multiple proteins involved in DNA replication, DNA repair, and cell cycle control bind to PCNA rather than directly associating with DNA, thus facilitating fast processing of DNA (reviewed in 3). PCNA protein expression is a well-accepted marker of proliferation. Applications: Suitable for use in Western Blot, Immunoprecipitation, Immunohistochemistry, Immunofluorescence/Immunocytochemistry and Flow Cytometry. Other applications not tested. Recommended Dilution: Western Blot: 1:1000, incubate membrane with diluted antibody in 5% BSA, 1X TBS, 0.1% Tween-20 at 4°C with gentle shaking, overnight. Immunoprecipitation: 1:50 Immunohistochemistry (Paraffin): 1:8000 Immunofluorescence (IF-IC): 1:800 Immunofluorescence (Frozen):1:800 Flow Cytometry: 1:100 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt.
