- Extra Details:
- Beta-catenin is one of the key downstream effectors in the Wnt signaling pathway (1). It is implicated in two major biological processes of vertebrates: early embryonic development (2) and tumorigenesis (3). Casein kinase 1 phosphorylates beta-catenin on Ser45. This phosphorylation event primes b-catenin for subsequent phosphorylation by GSK-3 (4-6). GSK-3b destabilizes b-catenin by phosphorylating it at Ser33, 37 and Thr41 (7). Mutations in these phosphorylation sites which result in the stabilization of beta-catenin protein levels have been found in many tumor cell lines (8). Applications: Suitable for use in Western Blot, Immunoprecipitation, Immunohistochemistry and ChIP. Other applications not tested. Recommended Dilution: Western Blot: 1:1000, incubate membrane with diluted antibody in 5% BSA, 1X TBS, 0.1% Tween-20 at 4°C with gentle shaking, overnight. Immunoprecipitation: 1:50 Immunohistochemistry (Paraffin): 1:1600 Immunohistochemistry (Frozen): 1:400 ChIP: 1:25 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
