- In eukaryotic cells, most proteins in the cytosol and nucleus are degraded via the ubiquitin-proteasome pathway. The 26S proteasome is a 2.5MD molecular machine built from approximately 31 different subunits, which catalyzes protein degradation. It contains a barrel-shaped proteolytic core complex (the 20S proteasome), capped at one or both ends by 19S regulatory complexes, which recognize ubiquitinated proteins. The regulatory complexes are also implicated in unfolding and translocation of ubiquitinated targets into the interior of the 20S complex, where they are degraded to oligopeptides. Applications: Suitable for use in Immunofluorescence/Immunocytochemistry and Western Blot. Not suitable for immunoprecipitation. Other applications not tested. Recommended Dilution: Western Blot: 1:100 Optimal dilutions to be determined by the researcher. Reactivity on Cultured Cell Lines: XLKE-A6 (Xenopus laevis), PLC (human), RVF (rat), Ptk2 (rat, kangaroo). Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
