- Human leucocyte antigen (HLA) class II molecules HLA-DP, HLA-DQ and HLA-DR are heterodimeric cell surface glycoproteins comprised of an alpha (heavy) chain and a beta (light) chain. They are expressed on B cells, activated T cells, monocytes/macrophages, dendritic cells, and other non-professional antigen presenting cells (APCs). HLA-Class II expressing APCs specifically present antigens recognized by TCR/CD3 complex of CD4+ T cells. Reactive microglia and perivascular cells in multiple sclerosis lesions express all three types of HLA-Class II molecules, whereas microglia in the normal CNS expressed HLA-DR only. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilution: Flow Cytometry: ≤0.5ug/1x10e6 cells in 100ul of staining volume or 0.1ml whole blood followed by a secondary florescent conjugated anti-mouse antibody Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
