- Oh et al. (1997) cloned mouse Spin from a 2-cell stage embryo library and showed that it is homologous to the multicopy Ssty gene. In situ hybridization and RT-PCR assays detected murine Spin transcript in unfertilized eggs and 2-cell stage embryos, but not in 8-cell stage embryos. Using differential display analysis of normal and cancerous ovarian tissues, followed by database analysis and PCR of total RNA from an ovarian cell line, Gao et al. (2005) cloned human SPIN. The 237aa protein has a predicted molecular mass of 27kD and shares 98% homology with mouse Spin. The C-terminal portion of SPIN was required for nuclear localization in transfected COS-7 cells. Applications: Suitable for use in ELISA, Western Blot, Immunohistochemistry, Immunocytochemistry. Other applications not tested. Recommended Dilution: ELISA: 1:100-1:5000 Western Blot: 1:50-400 Immunohistochemistry (frozen): 1:50-500 Immunohistochemistry (paraffin): 1:10-100 Immunocytochemistry: 1:50-500 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
