- Extra Details:
- The deduced NSHC sequence encodes isoforms of 474 and 594aa, corresponding to proteins of 52 and 64kD, respectively. Sequence analysis determined that NSHC shares an overall homology of 54% with SHC and approximately 70% homology in the PTB and SH2 domains. Northern blot analysis detected a 10-kb NSHC transcript exclusively in brain, whereas SHC was expressed as 3.8-, 3.4-, and 2.8-kb transcripts in all tissues except brain. Within the central nervous system, NSHC was highly expressed in cerebral cortex, hippocampus, amygdala, and caudate nucleus, with low-level expression in subthalamic nucleus, thalamus, putamen, corpus callosum, hypothalamus, medulla, substantia nigra, and cerebellum. Expression of NSHC was undetectable in the spinal cord. Applications: Suitable for use in ELISA, Western Blot, Immunohistochemistry, Immunocytochemistry. Other applications not tested. Recommended Dilution: Western Blot: 1:50-400 Immunocytochemistry: 1:50-500 Immunohistochemistry (frozen): 1:50-500 Immunohistochemistry (paraffin): 1:10-100 ELISA: 1:100-1:5000 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
