- Src homology 2 (SH2) domains bind specifically to tyrosine-phosphorylated proteins involved in growth factor and cytokine receptor signaling. The PTB domain of SHC1 specifically binds to the tyrosine-phosphorylated, 145kD protein INPP5D. By screening placental and hepatoma cDNA libraries using an EST encoding a putative PTB domain-containing protein as probe, Kavanaugh and Williams (1994) identified a partial cDNA encoding SCK (SHC-like). They determined that the SCK protein contains both a PTB and an SH2 domain. Northern blot analysis indicated that unlike SHC1, which is ubiquitously expressed with very low expression in brain, SCK is expressed higher in liver than in other tissues and is expressed in brain. Applications: Suitable for use in ELISA, Western Blot, Immunohistochemistry, Immunocytochemistry. Other applications not tested. Recommended Dilution: Western Blot: 1:50-400 Immunocytochemistry: 1:50-500 Immunohistochemistry (frozen): 1:50-500 Immunohistochemistry (paraffin): 1:10-100 ELISA: 1:100-1:5000 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
