- Elastase 4 is a member of the peptidase S1 family. The encoded protein is a serum calcium-decreasing factor that has chymotrypsin-like protease activity. Alternatively spliced transcript variants have been observed, but their full-length nature has not been determined. These human caldecrin isoforms, like the rat preproenzyme, are composed of a signal peptide of 16aa, a propeptide of 13aa, and a mature form of 239aa. Both recombinant caldecrins, produced in a baculovirus expression system, showed the same chymotrypsin-type protease activity and hypocalcemic activity. The hypocalcemic activity of both caldecrin isoforms remained intact even after treatment with phenylmethylsulfonyl fluoride (PMSF) to abolish their protease activity. Applications: Suitable for use in ELISA, Western Blot, Immunohistochemistry, Immunocytochemistry. Other applications not tested. Recommended Dilution: Western Blot: 1:50-400 Immunocytochemistry: 1:50-500 Immunohistochemistry (frozen): 1:50-500 Immunohistochemistry (paraffin): 1:10-100 ELISA: 1:100-1:5000 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
