- TOX (thymocyte selection-associated high mobility group (HMG) box protein) is a member of the HMG box family of DNA-binding proteins and likely plays a role in the regulation of T-cell development. It is a 526aa nuclear protein and the expression of TOX is upregulated by pre-T cell receptor (pre-TCR) and TCR activation in immature thymocytes. TOX-mediated positive selection is associated with upregulation of RUNX3 and is calcineurin dependent. TOX-dependent transition to the CD4+CD8 stage is required for development of class II major histocompatibility complex-specific T cells. Applications: Suitable for use in Immunofluorescence, ELISA and Western Blot. Other applications not tested. Recommended Dilution: Immunofluorescence: 20ug/ml Western Blot: 1-2ug/ml Optimal dilutions to be determined by the researcher. Positive Control: Human Colon Tissue Lysate Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
