- Extra Details:
- b-Catenin is a key downstream effector in the Wnt signaling pathway (1). It is implicated in two major biological processes in vertebrates: early embryonic development (2) and tumorigenesis (3). CK1 phosphorylates b-catenin at Ser45. This phosphorylation event primes beta-catenin for subsequent phosphorylation by GSK-3beta (4-6). GSK-3b destabilizes b-Catenin by phosphorylating it at Ser33, Ser37, and Thr41 (7). Mutations at these sites result in the stabilization of b-Catenin protein levels and have been found in many tumor cell lines (8). Applications: Suitable for use in Immunofluorescence, Flow Cytometry, Western Blot, Immunoprecipitation and Immunohistochemistry. Other applications not tested. Recommended Dilution: Immunofluorescence (frozen): 1:1600 Immunofluorescence (Immunocytochemistry): 1:1600 Flow Cytometry: 1:200 Western Blot: 1:1000, incubate membrane with diluted primary antibody in 5% BSA, 1X TBS, 0.1% Tween-20 at 4°C with gentle shaking, overnight. Immunohistochemistry (paraffin): 1:1000 Immunoprecipitation: 1:50 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
