- The matrix metalloproteinases (MMPs) are a large family of zinc endopeptidases that degrade extracellular matrix components such as fibronectin and collagen type III. The tissue inhibitors of metalloproteinase (TIMPs) complex with these matrix metalloproteinases and irreversibly inactivate them. The TIMP proteins are homologous and are either secreted in soluble form (TIMP1, TIMP2 and TIMP4) or bind to extracellular matrix components (TIMP3). Metalloproteinase inhibitor 4 (TIMP4) is known to inhibit MMP-1, MMP-2, MMP-3, MMP-7 and MMP-9. Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 1:100-1000 Immunohistochemistry: 1:100-200 Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
