- Tim-2 (T-cell immunoglobulin mucin 2), a transmembrane glycoprotein and member of the immunoglobulin superfamily, is expressed by activated lymphocytes, developmentally in the 8-cell embryo and blastocyst, and preferentially up-regulated in Th2 cells and down-regulated in Th1 cells. Tim-2 acts as a negative regulator of Th2 cells, and is implicated to play a critical role in the development of atopic disease and other Th2-biased immune responses. Tim-2 also acts as a receptor for Semaphorin 4A, a transmembrane protein expressed on dendritic cells and B cells, which plays a role in the guidance of axonal migration during neuronal development, and has been shown to enhance T cell activation. Studies using TIM-2 in mouse have shown that signaling by Tim-2 can influence the proliferation and antibody production of B cells, during early phase CIA (collagen-induced arthritis) (Kawamoto et al. 2011). TIM-2 has been shown to significantly enhance the proliferation of stimulated splenic B cells in vitro. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1:500-1:1000 Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
