- This product recognizes both the pro and active form of MMP-10. The MMP’s are zinc-dependent endopeptidases responsible for degrading the extracellular matrix. They are also involved in cell proliferation, migration, differentiation and apoptosis. Most MMP’s are synthesised as inactive zymogens and a propeptide region must be cleaved off before the enzyme becomes active. Their expression is increased dramatically in a variety of cancer types, where it indicates invasive disease and a poor prognosis. MMP-10 is a collagenase capable of degrading fibronectin, gelitins (types I, III, IV and V), and more weakly collagens III, IV and V. MMP-10 activates procollagenase. Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 1:1000 Immunohistochemistry: 1:50-300 Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
