- GITR, glucocorticoid-induced TNFR-related gene, is a member of the TNF receptor superfamily, also known as TNFRSF18, and AITR (in humans). It is expressed at low levels on resting T lymphocytes and at high levels on CD25+CD4+Treg cells. The expression of GITR on T cells can be upregulated upon activation. Interaction of GITR with its ligand (GITRL) has been demonstrated to augment T cell activation, proliferation, cytokine production, as well as MAPKs and NF-kB activation, and abrogate the inhibitory function of CD25+CD4+ T reg cells. In vivo activation of GITR causes development of autoimmune diseases and restores the suppressed immune response. Applications: Suitable for use in Flow Cytometry and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Flow Cytometry: 1:50-1:1000 Immunohistochemistry (Paraffin): 1:50-1:200 Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
