- alpha1-antitrypsin (AAT) is a protective protease inhibitor. It inhibits elastase released by neutrolphils during inflammatory reaction and irreversibly inactivates trypsin secreted in duodenum and elsewhere. alpha1-antitrypsin normally presents in plasma at 1.5-3.5mg/ml, but increases in acute phase reaction, malignancies, pregnancy and trauma. Severe hereditary alpha1-antitrypsin deficiency can result in damages to alveoli, chronic obstructive pulmonary disease (COPD), and liver diseases such as jaundice and cirrhosis. The protein is also found in the neurofibrillary tangles and senile plaques in the brain lesions of Alzheimer’s disease, and has been suggested to contribute to the pathology of Alzheimer’s disease. Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1:5000 Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
