- Siglec-10 (Sialic acid-binding Ig-like lectin 10), a putative adhesion molecule and member of the Ig superfamily, is expressed by monocytes, B cells, eosinophils, and at a higher level by a subpopulation of CD16+CD56- natural killer (NK) cells. Structurally, Siglec-10 is most similar to the CD33-related group of Siglecs, and preferentially binds to glycoconjugates containing alpha-2,3- or alpha-2,6-linked sialic acid. Studies have shown that Siglec-10 acts as a substrate for VAP-1 (Vascular adhesion protein-1), a glycoprotein expressed on endothelium during inflammation, which is involved in primary amine oxidation and leucocyte trafficking, see Kive, E. et al. for details. This interaction between Siglec-10 and VAP-1, implicates Siglec-10 in endothelial lymphocyte adhesion and in the modulation of the inflammatory microenvironment. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilution: Flow Cytometry: 10ul labels 10e6 cells Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: APC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Note: Applications are based on unconjugated antibody.
