- Extra Details:
- Lipoprotein lipase (LPL) is a member of the AB hydrolase superfamily, which plays a pivotal role in lipoprotein metabolism and transport, acting as the key enzyme in the hydrolysis of triglycerides and very low density lipoproteins (VLDLs), and the release of free fatty acids into peripheral tissues. A deficiency of LPL can result in hypertriglyceridemia, and many studies have focused on the critical role which LPL plays in the pathogenesis of atherosclerosis, and in particular the relationship between LPL and apolipoprotein E (ApoE), both of which are secreted in significant amounts by macrophages in developing arterial wall lesions. 214981 is a unique antibody which differentiates between monomeric inactive and dimeric active LPL, and binds to LPL sequences involved in LPL, LPL receptor, and heparin interactions. 214981 has been shown to inhibit the activity of human LPL (Chang et al. 1998), and is widely used in human studies. Applications: Suitable for use in ELISA, Western Blot, Immunoprecipitation. Other applications not tested. Recommended Dilution: ELISA: 1:50-1:200 in a direct ELISA, or as the detection reagent in a sandwich ELISA with 214978 as the capture antibody. Western Blot: detects a band of ~53kD using partially purified LPL from postheparin plasma. Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
