- Extra Details:
- IL-10 is an immunosuppressive cytokine produced by a variety of mammalian cell types including macrophages, monocytes, T cells, B cells and keratinocytes. IL-10 inhibits the expression of proinflammatory cytokines such as IL-1 and TNF-α. Like IL-4, IL-10 enhances humoral immune responses and attenuates cell-mediated immune reactions. Human IL-10 is active on murine cells, but murine IL-10 is inactive on human cells. Recombinant human IL-10 is an 18.6kD protein of 161aa residues. Recombinant protein corresponding to human IL-10, expressed in E. coli. Accession/Uniprot: P22301 Amino Acid Sequence: MSPGQGTQSE NSCTHFPGNL PNMLRDLRDA FSRVKTFFQM KDQLDNLLLK ESLLEDFKGY LGCQALSEMI QFYLEEVMPQ AENQDPDIKA HVNSLGENLK TLRLRLRRCH RFLPCENKSK AVEQVKNAFN KLQEKGIYKA MSEFDIFINY IEAYMTMKIR N Biological Activity: Determined by the dose-dependent co-stimulation (with murine IL-4) of MC/9 cells. The ED50 was found to be ≤ 2ng/ml, corresponding to a specific activity of ≥ 5x10e5 units/mg. Storage and Stability: Lyophilized and reconstituted products are stable for 6 months after receipt at -20°C. Reconstitute with sterile 5mM Tris, pH 7.2. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
