- The formation of free radicals and other highly reactive oxygen species has been implicated in the pathogenesis of many disease states. The ability to identify these species is crucial, and spin trapping has accomplished this goal. DMPO (5,5-dimethyl-1-pyrroline N-oxide) is one of the least toxic to cells and animals, and possesses convenient pharmacokinetics (uptake, distribution, metabolism and excretion) in biological systems. Recent studies have determined that nitric oxide may substantially affect the quantitative determination of DMPO adducts, and therefore extra caution is required when studying generation of these species in the presence of nitric oxide or its radicals. DMPO adducts can be generated with protein and DNA radicals. Applications: Suitable for use in Immunofluorescence, ELISA, Western Blot, Immunoprecipitation and Immunocytochemistry. Other applications not tested. Recommended Dilution: ELISA: 1-10ug/ml Immunocytochemistry: 10ug/ml Immunoprecipitation: 25ug Optimal dilutions to be determined by the researcher. Extinction Coefficient: 90,000 Excitation (Max): 501nm Emission (Max): 523nm Storage and Stability: May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and store at -20°C. Aliquots are stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. ATTO 488 conjugates are sensitive to light.
