- This MAb recognizes basic (Type II or HMW) cytokeratins, which include 67kD (CK1); 64kD (CK3); 59kD (CK4); 58kD (CK5); 56kD (CK6); 52kD (CK8). Twenty human keratins are resolved with two-dimensional gel electrophoresis into acidic (pI <5.7) and basic (pI >6.0) subfamilies. The acidic keratins have molecular weights (MW) of 56.5, 55, 51, 50, 50’, 48, 46, 45, and 40kD. This MAb recognizes the 65-67, 64, 59, 58, 56, and 52kD keratins of basic subfamily. Many studies have shown the usefulness of keratins as markers in cancer research and tumor diagnosis. It is a broad spectrum anti pan-keratin antibody cocktail, which differentiates epithelial tumors from non-epithelial tumors e.g. squamous vs. adenocarcinoma of the lung, liver carcinoma, breast cancer, and esophageal cancer. Applications: Suitable for use in Immunofluorescence, Flow Cytometry, ELISA, Western Blot, Immunohistochemistry. Other applications not tested. Recommended Dilution: Flow Cytometry: 0.5-1ug/million cells Immunofluorescence: 1-2ug/ml Western Blot: 0.5-1ug/ml Immunohistochemistry (Frozen & Formalin-fixed): 0.5-1ug/ml for 30 min at RT (Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 minutes) Optimal dilution for a specific application should be determined. Positive Control: Skin, Adenocarcinomas Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
