- Actin is a highly conserved ubiquitous globular protein (G-actin) that polymerizes to form fibrous F-actin microfilaments. In higher eucaryotes several actin isoforms have been identified, that fall into three classes. Alpha actin is a structural component of the contractile apparatus of muscle cells or muscle-derived cells. Beta actin and gamma actin play roles in regulation of cell motility in other cell types. Specific subcellular structures such as stress fibers, focal adhesions, filopodia etc., are formed by involvement of actin cytoskeleton. Applications: Suitable for use in Western Blot, Immunohistochemistry and Immunocytochemistry. Other applications not tested. Recommended Dilutions: Western Blot: 1ug/ml using murine femoral muscle, murine heart as positive controls, and HUVEC line as a negative control under reducing conditions. Immunohistochemistry (paraffin sections): Antigen retrieval steps generally not required, but e.g. in case of arterial smooth muscle cells or myoepithelial cells, pepsin or trypsin pretreatment is recommended. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
