- CD6 is a type I transmembrane glycoprotein that contains a 24-amino acid signal sequence, three extracellular “scavenger receptor cysteine-rich” (SRCR) domains, a membrane-spanning domain and a 44-amino acid cytoplasmic domain. The CD6 glycoprotein is tyrosine phosphorylated during TCR-mediated T cell activation. CD6 shows significant homology to CD5. CD6 is present on mature thymocytes, peripheral T cells and a subset of B cells. Antibodies to CD6 are used to deplete T cells from bone marrow transplants to prevent graft versus host disease. Applications: Suitable for use in Immunofluorescence, Flow Cytometry, Western Blot, Immunoprecipitation, Immunohistochemistry. Other applications not tested. Recommended Dilution: Flow Cytometry: 0.5-1ug/million cells Immunofluorescence: 0.5-1ug/ml Western Blot: 0.5-1ug/ml Immunoprecipitation: 0.5-1ug/500ug protein lysate Immunohistochemistry: Frozen & Formalin-fixed: 0.5-1ug/ml for 30 minutes at RT (Staining of formalin-fixed tissues requires boiling tissue sections in 10mM Citrate Buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 minutes Optimal dilutions to be determined by the researcher. Positive Control: CCRF-CEM, Jurkat cells, Tonsil Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
