- Signal transducer and activator of transcription 1 (Stat1) belongs to a family of cytoplasmic transcription factors that can be activated (phosphorylated) by a cell surface receptor. Phosphorylation of Stat1 at 701aa induces Stat1 dimerization, nuclear translocation and DNA binding. Stat1 has two isoforms, Stat1a (91kD) and the splice variant Stat1b (84kD). Stat1 has been found to be inappropriately activated in many tumors. In addition to tyrosine phosphorylation, Stat1 is phosphorylated through a p38 mitogen-activated protein kinase (MAPK)-dependent pathway at Ser727 in response to IFN-g and other cellular stresses. Stat1 is localized in the cytoplasm and upon phosphorylation at Y701 is translocated to the nucleus. Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilution: ELISA: 1:25,000 Western Blot: 1:500-1:2000 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
