Extra Details:: IL-3 is a pleiotropic cytokine that can stimulate proliferation and differentiation of pluripotent hematopoietic stem cells as well as various lineage committed progenitors.1, 2 1L-3 exerts its activity through binding to a specific cell surface receptor known as IL-3 R. IL-3 R is a heterodimeric structure composed of a 70kD IL-3 Ra subunit (CDw123 or beta common) and a 120-140kD IL-3 Rb subunit (CDw131).3, 4 IL-3 Ra binds IL-3 with relatively low affinity. In the presence of IL-3 Rb however, IL-3 Ra has a much higher affinity for IL-3. It is not clear how signal transduction occurs following IL-3 binding. The IL-3 Ra chain has a very short intracellular domain while the IL-3 Rb chain has a very large cytoplasmic domain. Recent studies suggest that formation of b-b homodimers initiates signaling events.5 The IL-3 Rb chain is also shared by the receptors for IL-5 and GM-CSF. Cells known to express IL-3 receptors include hematopoietic progenitors, epithelial cells, double negative T cells, mast cells, basophils, and blood monocytes. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilution: Flow Cytometry: 10ul/10e6 cells. Sampled with human peripheral blood lymphocytes Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4°C. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Allophycocyanin (APC) is light sensitive.
