Extra Details:
Platelet Receptor Gi24, also known as VISTA, B7H5, B7H5, Dies1 (Differentiation of ESC1), SISP1 and C10orf54, is a 5565kD member of the Ig superfamily. It is a transmembrane molecule expressed in bone, on embryonic stem cells (ESCs), and on tumor cell surfaces. On ESCs, Gi24 appears to positively interact with BMP4, potentiating BMP signaling and the transition from an undifferentiated to a differentiated state. On tumor cells, Gi24 both promotes MT1MMP expression and activity, and serves as a substrate for MT1MMP. This increases the potential for cell motility. Mature mouse Gi24 is a 277 amino acid (aa) type I transmembrane glycoprotein (aa33309). It contains a 149aa extracellular region (aa33191) with one Vtype Iglike domain (aa33161) and a 97aa cytoplasmic domain. Based on human Gi24, mouse Gi24 will likely undergo proteolytic cleavage by MT1MMP, generating a soluble 30kD extracellular fragment, plus a 2530kD membranebound fragment. There are two potential isoform variants. One contains a deletion of aa127187, while another shows an alternative start site at aa82. Over aa33191, mouse Gi24/Dies1 shares 78% and 70% aa identity with rat and human Gi24, respectively. Applications: Suitable for use in ELISA, Western Blot and Immunocytochemistry. Other applications not tested. Recommended Dilutions: Western Blot: 2ug/ml using mouse platelets Immunocytochemistry: 8-25ug/ml using immersion fixed D3 mouse embryonic stem cell line Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
