- MYF5 (Myogenic factor 5; also bHLHc2) is a 3739kD transcriptional activator that belongs to the bHLH superfamily of myogenic regulatory factors. It is found in embryonic skeletal muscle progenitors and neurons, and is induced by SHH and Wnt1. Expression of MYF5 protein prompts nonmuscle cells to adopt a muscle precursor phenotype. Notably, neurons appear to block MYF5 accumulation, thus preserving their unique phenotype. MYF5 forms heterodimers with multiple molecules. When complexed to Imfa, MYF5 is retained in the cytoplasm and is inactive; when complexed to E12, MYF5 is transcriptionally active. Phosphorylation regulates MYF5 stability. Human MYF5 is 255 amino acids (aa) in length. It contains one myogenic basic region (aa183) followed by an HLH motif (aa96135). Fulllength human MYF5 (aa1255) shares 89% aa sequence identity with mouse MYF5. Applications: Suitable for use in ELISA and Immunocytochemistry. Other applications not tested. Recommended Dilution: Immunocytochemistry: 825ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
