Extra Details:: Lefty2 (Leftright determination factor 2; also LeftyB [in human]) is a atypical member of the TGFbeta family of proteins. It is expressed during early embryogenesis in both the primitive streak and leftside lateral plate mesoderm. In the adult, Lefty2 appears in oviduct epithelium. Lefty2 acts in a manner reminiscent of that for Chordin and Noggin, and it is assumed that Lefty2 is an antagonist of BMP activity. Notably, Lefty2 and Nodal are likely under the control of Lefty1, and thus all three molecules would appear to contribute to the creation of a left sidetype body plan. Mouse Lefty2 is synthesized as a 368 amino acid (aa) preproprecursor. It contains a 21aa signal sequence, plus a 347aa, 4142kD bioactive proprecursor that may undergo proteolytic processing at one of two downstream cleavage sites. If cleavage occurs after Arg77, the resulting 3334kD mature form (aa78368) is biologically inactive; if cleavage occurs after Arg135, the resulting 2728kD mature form (aa136368) is biologically active. Lefty2 is not a covalent homodimer and has been suggested to act as a monomer. Over aa78368, mouse Lefty2 shares 94%, 83% and 95% aa sequence identity with rat Lefty2, human LeftyB and mouse Lefty1, respectively. Applications: Suitable for use in ELISA and Immunocytochemistry. Other applications not tested. Recommended Dilution: Immunocytochemistry: 825ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
