- Complexin1 (CPX I; also synaphin2) is a 1718kD, cytosolic protein that belongs to the complexin/synaphin family, complexin1/2 subfamily of molecules. It is expressed in select neurons and pancreatic betacells, and serves as a block against premature neurotransmitter/insulin granule release. Both Complexin1 and Complexin2 bind to the SNARE complex that contains SNAP25, VAMP2 and STX1A, generating oligomers of SNARE complexes that are optimally oriented relative to the positions of vesicle and plasma membranes. This effectively places synaptic vesicles in an optimal configuration, allowing for a coordinated, uniform fusion and release upon appropriate stimulus. Typically, complexin1 is described as a clamp that blocks (vesicle) fusion, and the stimulus that overcomes it is one that induces an influx of synaptotagmindetectable calcium. Human complexin1 is 134 amino acids (aa) in length. It contains one coiledcoil domain (aa2969) that incorporates part of a SNARE complex interaction sequence (aa4870). Fulllength human complexin1 shares 97% aa sequence identity with mouse complexin1. Applications: Suitable for use in ELISA, Western Blot, and Immunocytochemistry. Other applications not tested. Recommended Dilution: Western Blot: 0.5ug/ml Immunocytochemistry: 515ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile ddH2O or PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
