- Extra Details:
- NCAM-1 (Neural adhesion molecule1; also known as CD56) is a 120-190kD glycoprotein member of the Ig superfamily. It is expressed on multiple cell types, both in the embryo and adult. It serves as both an adhesion molecule and a receptor for multiple ligands, including FGF, PDGF, GDNF, and Agrin. On the cell surface, it is a cis-oriented homodimer that can form homodimers in trans with other cis homodimers. Over amino acids 20-711, mouse NCAM-1 shares 99% and 95% amino acid sequence identity with rat and human NCAM-1, respectively. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilutions: Flow Cytometry: 0.25-1ug/10e6 cells. Using Neuro-2A mouse neuroblastoma cell line Optimal dilutions to be determined by the researcher. Flow Cytometry: This antibody has been tested for flow cytometry using mouse Neuro2A cells. 1. Cells may be Fc-blocked with 1ug of mouse IgG/10e5 cells for 15 minutes at room temperature. Do not wash excess blocking IgG from this reaction. 2. After blocking, 10ul of 145420 was added to up to 1x10e6 cells and incubated for 30 minutes at room temperature. 3. Unbound antibody was removed by washing the cells twice in F4568-10: Flow Cytometry Staining Buffer, 1X. Note that whole blood requires a RBC lysis step at this point using Flow Cytometry Mouse Lyse Buffer. 4. The cells were resuspended in F4568-10: Flow Cytometry Staining Buffer, 1X for final flow cytometric analysis. As a control for this analysis, cells in a separate tube should be treated with APC-labeled rat IgG2A antibody. Storage and Stability: Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: APC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
