- Bcl2 antagonist of cell death (Bad) is a 23kD cytoplasmic protein in the Bcl2 family. It functions as a proapoptotic molecule by dimerizing with and inhibiting the antiapoptotic proteins Bcl2 and BclxL. Prosurvival signals trigger the phosphorylation of Bad on Ser115, disrupting its interaction with Bcl2 and BclxL and resulting in protection from apoptosis. Human Bad shares 75% aa sequence identity with mouse and rat Bad. Applications: Suitable for use in ELISA, Western Blot, and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 0.5ug/ml Immunohistochemistry: 825ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile ddH2O or PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
