- 1433 proteins are a highly conserved family of homo and heterodimeric phosphoserine/threoninebinding proteins present in high abundance in all eukaryotic cells. 1433 proteins were the first polypeptides shown to have pSer/Thr binding properties, generally recognizing the consensus sequences RSXpSXP and RXY/FXpSXP (where X is any amino acid). 1433 proteins act as key regulators of intracellular signal transduction through their ability to bind specific motifs phosphorylated on serine or threonine. For example, the binding of 1433 to phosphorylated BAD blocks its proapoptotic association with BclXL. There are at least seven distinct 1433 genes in vertebrates, alpha/beta, epsilon, eta, gamma, theta, sigma and zeta. 1433 zeta, also known as Tyrosine 3 Monooxygenase/Tryptophan 5Monooxygenase Activation Protein, zeta isoform (gene name YWHAZ) is a 245 amino acid, 27kD protein. Applications: Suitable for use in ELISA, Western Blot, and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 0.2ug/ml Immunohistochemistry: 825ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
