Extra Details:
Growth factor active in angiogenesis, vasculogenesis and endothelial cell growth. Induces endothelial cell proliferation, promotes cell migration, inhibits apoptosis and induces permeabilization of blood vessels. Binds to the FLT1/VEGFR1 and KDR/VEGFR2 receptors, heparan sulfate and heparin. NRP1/Neuropilin-1 binds isoforms VEGF-165 and VEGF-145. Isoform VEGF165B binds to KDR but does not activate downstream signaling pathways, does not activate angiogenesis and inhibits tumor growth. Applications: Suitable for use in FLISA, Western Blot and Neutralization. Other applications not tested. Recommended Dilution: FLISA: In a sandwich FLISA (assuming 100ul/well), a concentration of 2-4ug/ml of this antibody will detect at least 100pg/ml of recombinant human VEGF165 when used with biotinylated antigen affinity purified anti-human VEGF165 as the detection antibody at a concentration of approximately 0.25-1ug/ml. Western Blot: To detect hVEGF165 by Western Blot analysis this antibody can be used at a concentration of 0.25-0.5ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hVEGF165 is 0.25-0.5ng/lane, under non-reducing conditions. Neutralization: To yield one-half maximal inhibition of the biological activity of Human VEGF (10ng/ml), a concentration of 0.2ug/ml of this antibody is required. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C before opening. DO NOT FREEZE! Stable at 4°C as an undiluted liquid. Dilute only prior to immediate use. Stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer. Allophycocyanin (APC) is light sensitive. Note: Applications are based on unconjugated antibody.
