- Cytokine with immunoregulatory activity. May promote the transition between innate and adaptive immunity. Induces the production of IgG(1) and IgG(3) in B-cells (By similarity). May play a role in proliferation and maturation of natural killer (NK) cells in synergy with IL15. May regulate proliferation of mature B-and T-cells in response to activating stimuli. In synergy with IL15 and IL18 stimulates interferon gamma production in T-cells and NK cells. During T-cell mediated immune response may inhibit dendritic cells (DC) activation and maturation. Applications: Suitable for use in ELISA, Western Blot and Neutralization. Other applications not tested. Recommended Dilutions: ELISA: To detect hIL-21 by sandwich ELISA (using 100ul/well antibody solution) a concentration of 0.5-2ug/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with Biotinylated Anti-Human IL-21 as a detection antibody, allows the detection of at least 0.2-0.4ng/well of recombinant hIL-21. Western Blot: To detect hIL-21 by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hIL-21 is 1.5-3ng/lane, under either reducing or non-reducing conditions. Neutralization: This antibody exhibits complete neutralization of the biological activity of hIL-21 in a human B cell stimulation assay. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
