- MaxLight™490 is a new Blue-Green photostable dye conjugate comparable to DyLight™488, Alexa Fluor™488 and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (491nm); Emission (515nm); Extinction Coefficient 73,000. Produced by T-cells in response to antigenic or mitogenic stimulation, this protein is required for T-cell proliferation and other activities crucial to regulation of the immune response. Can stimulate B-cells, monocytes, lymphokine-activated killer cells, natural killer cells, and glioma cells. Applications: Suitable for use in FLISA, Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: FLISA: In a sandwich FLISA (assuming 100ul/well), a concentration of 4-8ug/ml of this antibody will detect at least 1000pg/ml of recombinant human IL-2 when used with biotinylated antigen affinity purified anti-human IL-2 as the detection antibody at a concentration of approximately 1-2ug/ml. Western Blot: To detect hIL-2 by Western Blot analysis this antibody can be used at a concentration of 1-2ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hIL-2 is 2-4ng/lane, under reducing or non-reducing conditions. Immunohistochemistry: This antibody stained formalin-fixed, paraffin-embedded sections of human breast invasive ductal carcinoma. The recommended concentration is 2.5ug/ml-5ug/ml with an overnight incubation at 4°C. An HRP-labeled polymer detection system was used with a DAB chromogen. Heat induced antigen retrieval with a pH 6.0 sodium citrate buffer is recommended. Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™490 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Note: Applications are based on unconjugated antibody.
