- Interferons (IFNs) are proteins made and released by lymphocytes in response to the presence of pathogens-such as viruses, bacteria, or parasites-or tumor cells. They allow communication between cells to trigger the protective defenses of the immune system that eradicate pathogens or tumors. Applications: Suitable for use in Neutralization, ELISA and Western Blot. Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. Activity: > 5x10e3 neutralizing units/mg Bioactivity Assay: One neutralization unit is the amount of antiserum required to neutralize one unit of recombinant rat interferon beta (rat IFN-β) to a 50% endpoint (one unit corresponds to approximately 15pg of purified rat IFN-β). Interferon was titrated with the use of the cytopathic effect inhibition assay as described [Rubinstein, S., Familletti, P.C., and Pestka, S. (1981) "Convenient Assay for Interferons," J. Virol. 37, 755-758; Familletti, P.C., Rubinstein, S., and Pestka, S. (1981) "A Convenient and Rapid Cytopathic Effect Inhibition Assay for Interferon," in Methods in Enzymology, Vol. 78 (S. Pestka, ed.), Academic Press, New York, 387-394]. In this antiviral assay for interferon about 1 unit/ml of interferon is the quantity necessary to produce a cytopathic effect of 50%. As there is no international rat IFN-β standard, the units are determined with respect to the international reference standard for mouse interferon beta (mouse IFN-β) provided by the National Institutes of Health [see Pestka, S. (1986) "Interferon Standards and General Abbreviations," in Methods in Enzymology (S. Pestka, ed.), Academic Press, New York 119, 14-23]. This material is prepared specifically for effective neutralization of rat IFN-β. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
