- Tissues are continually exposed to reactive oxygen species (ROS) that are produced in tissues from metabolism processes or the inflammatory response of leukocytes or macrophages. These ROS can cause damage to various cellular components. Dinitrophenylhydrazine (DNPH) will react with free carbonyl groups on proteins, leading to adduction of the dinitrophenyl (DNP) group to the carbonyl group. Using an antibody to DNP, it is possible to visualize and localize the sites of oxidative damage by immunodetection of DNP adduction resulting from reaction of the tissue with DNPH. This anitbody has been used in DNA immunoFISH and for differential staining of blastocysts. Was used to study the effects of IgE on the clustering of FcεRI by time-resolved phosphorescence anisotropy. Applications: Suitable for use in Indirect ELISA and Quantitative Precipitin Assays. Other applications not tested. Recommended Dilution: Indirect ELISA: 1:8000. Quantitative Precipitin Assays: 1.6-2.4mg/ml. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
