- GTP-binding protein Rit2 (Rit2; also Rin, Ras-like protein expressed in neurons, Ras-like without CAAX) is a 25kD member of the small GTPase superfamily and Ras family. Human Rit2 is 217aa in length. Like all small G-proteins, Rit2 contains five highly conserved domains (G1-G5) and acts as a molecular switch by alternating an active GTP-bound form and an inactive GDP-bound form. There are two isoforms for Rit2. Isoform 1 is the long form. Isoform 2 has an 11aa substitution for residues 143-153 found in isoform 1 and a deletion of aa154-217. Rit2 is expressed exclusively in neurons. In addition, Rit2 binds calmodulin through a C-terminal binding motif. Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
