- The MBD family of proteins includes MeCP2, MBD1, MBD2, MBD3 and MBD4. MBD1 (methyl-CpG binding domain protein 1) binds to methylated CpG sites and represses gene transcription. MBD1 is recruited to both methylated and non-methylated CpGs by separate domains: the CXXC domain targets the protein to non-methylated CpGs whereas the MBD domain targets the protein to methylated CpG. MBD1 may also interact with histone methyltransferase SETDB1 to repress transcription. Applications: Suitable for use in ELISA, Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Immunohistochemistry (Formalin fixed paraffin embedded): 3ug/ml Optimal dilutions to be determined by the researcher. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
