Extra Details:: These chimeric antibodies are derived from mouse monoclonal AAV antibodies and are a combination of the mouse antigen binding region and a human Fc region. Therefore, it provides the well-known characteristics of the corresponding mouse monoclonal antibody (anti-AAV1, ADK1a, (cat # 553700)) like binding affinity, cross-reactivity and neutralization activity combined with a human Fc region allowing the use in an anti-human secondary antibody detection system. Many humans in the general population have developed antibodies against AAV as a result of naturally acquired infections, which might affect efficacy and safety of the gene transfer using AAV vectors. Therefore, testing for pre-existing AAV antibodies in patient sera is an indispensable step for the selection of patients for AAV gene therapy clinical trials. Human chimeric AAV antibodies are close to the human derived samples, making them the ideal positive control for reproducible and comparable serological assays. Applications: Suitable for use in Dot Blot, Neutralization and Serology ELISA. Other applications not tested. Recommended Dilutions: Dot Blot: 1:500 (0.1ug/ml; non-denaturing conditions). Neutralization Assay: EC50 ~1ng/ml (AAV1) and ~1ng/ml (AAV6). Assay dependent. Serology Assay: 0.1-0.4ug/ml (1:125-1:500; AAV6 ELISA). Immunoprecipitation: 1:5 Optimal dilutions to be determined by the researcher. Binding Affinity: KD value (AAV1) = 8.8E-12 M KD value (AAV6) = 5.9E-11 M Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
