- Glypicans are GPI-anchored heparan sulfate proteoglycans that include a large globular cysteine-rich domain with 14 invariant cysteine residues, a stalk-like region containing the heparan sulfate attachment sites, and a C-terminal GPI attachment site. Glypicans are thought to function as low-affinity binding proteins that retain signaling molecules (cytokines, growth factors, proteases) at the cell surface. Glypicans are widely expressed in adult and fetal tissues. Applications: Suitable for use in ELISA, Flow Cytometry, Western Blot, Immunohistochemistry and Immunocytochemistry. Other applications not tested. Recommended Dilutions: Flow Cytometry: 2.5ug/10e6 cells using HepG2 human hepatocellular carcinoma cell line as a sample. Immunocytochemistry: 1-5ug/ml in immersion fixed HepG2 human hepatocellular carcinoma cell line Western Blot: 2ug/ml with HepG2 human hepatocellular carcinoma cell line as samples, under reducing conditions Immunohistochemistry: 5-15ug/ml. Using Immersion fixed paraffin-embedded sections of human breast and immersion fixed paraffin-embedded sections of human liver cancer tissue as samples. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
