- Extra Details:
- The CNS is enriched in polyunsaturated fatty acids and is therefore particularly vulnerable to lipid peroxidation. 4-Hydroxy-2-noneal (HNE) is one of the major lipid peroxidation products with cytotoxic and mutagenic activity. It reacts with protein residues such as histidine to generate stable Michael adducts. HNE has been shown to inhibit neurite outgrowth and disrupt microtubules and tubulin modification, suggesting it may play a role in Alzheimer′s disease. Note: Does not bind non-reduced adducts and reduced modifications arising from malondialdehyde (MDA) or structurally-related alkenals such as trans-2-noneal. Does not cross-react with non-reduced amino acid-HNE Michael adducts. Antibody binding requires the presence of the 4-hydroxyl group. Variables associated with assay conditions will dictate the optimal working dilution. Applications: Suitable for use in ELISA, Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilutions: ELISA: 1:500-1:1000 Western Blot: 1:1000-1:4000 Immunohistochemistry (frozen and paraffin): 1:50-1:500 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
